چکیده

Introduction: Hypertension has emerged as a serious adverse effect of immunosuppression with cyclosporin, which has become the mainstay of immunosuppression in organ transplantation. Improved survival rates with cyclosporin compared to previous regimens based on corticosteroids and azathioprine were established and have led to an expansion of solid organ transplantation. Cyclosporin has also been used at lower dosages for the treatment of autoimmune disease.

Vasomotor responses in cyclosporin A-treated rats after chronic angiotensin blockade.

Chronic angiotensin-converting enzyme (ACE) inhibition prevents endothelial dysfunction in hypertension and hypercholesterolemia. Long-term treatment with cyclosporin A impairs endothelium-dependent relaxations and augments contractions to angiotensin II in the rat aorta. The present study compares vasomotor responses to several vasoconstrictor and dilator stimuli after 6 weeks of oral treatmen...

NFATc3 is required for intermittent hypoxia-induced hypertension.

Sleep apnea, defined as intermittent respiratory arrest during sleep, is associated with increased incidence of hypertension and peripheral vascular disease. Exposure of rodents to brief periods of intermittent hypercarbia/hypoxia (H-IH) during sleep mimics the cyclical hypoxia-normoxia of sleep apnea. Endothelin-1, an upstream activator of nuclear factor of activated T cells (NFAT), is increas...

Cyclosporin A-Induced Hyperreninemic Hypoaldosteronism

We studied the effects of cyclosporin A on the renin-aldosterone axis in SpragueDawley rats. Two weeks of intragastric administration of cyclosporin A (5 mg/kg/day or 20 mg/kg/day) resulted in large increases in plasma renin concentration (23 ± 5, 70 ±12, and 79 ± 11 ng/ml/hr in control rats and rats receiving 5 mg and 20 mg of cyclosporin A, respectively), with no parallel increments in plasma...

Cyclosporin-induced dyslipoproteinemia is associated with selective activation of SREBP-2.

The use of cyclosporin A has contributed greatly to the success of organ transplantation. However, cyclosporin-associated side effects of hypertension, nephrotoxicity, and dyslipoproteinemia have tempered these benefits. Cyclosporin-induced dyslipoproteinemia may be an important risk factor for the accelerated atherosclerosis observed posttransplantation. Using a mouse model, we treated Swiss-W...

A Study of arbutin protective effect on cyclosporin A-induced oxidative damage

Background: Cyclosporine A (CsA) is a potent immunosuppressant drug with therapeutic and toxic actions. The use of CsA is limited by its toxicity. Several researchers had proposed that oxidative stress could play an important role in CsA-induced toxicity. Arbutin has recently been shown to possess antioxidative and free radical scavenging abilities. The present study was designed to investigate...
